Immunohistochemical staining using Collagen Type I antibody (biotin) (60R-CR004bt)

Tissue: Human Skin at pH6. Fixation: formalin fixed paraffin embedded. Collagen Type I antibody at 10 ug/mL for 1 h at RT. Peroxidase rabbit secondary antibody at 1:10,000 for 45 min at RT. Localization: Collagen Type I is secreted in the extracellular matrix. Staining: Collagen Type I as precipitated brown signal (A) with hematoxylin purple nuclear counterstain. With corresponding negative conrol (B).

Specifications

Host Rabbit
Method of Purification Collagen Type I antibody (biotin) was purified by affinity chromatography.
Tag/Conjugate Biotin
Form & Buffer Lyophilized from 0.125M Na3B4O7, pH 8.0, with 0.075M NaCl, 0.005M EDTA, 10 mg/ml BSA, and 0.01% NaN3. Immunoglobulin and protease free.

Usage & Assay Information

Usage Recommendations ELISA: 1:3,000-1:6,000, IHC: 1:50-1:200, WB: 1:3,000-1:6,000
Bioactivity Some class specific anti-collagens may be specific for three-dimensional epitopes which may result in diminished reactivity with denatured collagen or formalin-fixed, paraffin embedded tissues

Storage & Safety

Storage Store at 4 deg C until reconstitution. Following reconstitution aliquot and freeze at -20 deg C for long term storage. Avoid repeated freeze/thaw cycles.
Biohazard Information This product contains sodium azide as preservative. Although the amount of sodium azide is very small appropriate care must be taken when handling this product.

General Information

Biological Significance Collagen is a group of naturally occurring proteins found, in nature, exclusively in animals, especially in the flesh and connective tissues of mammals. It is the main component of connective tissue, and is the most abundant protein in mammals, making up about 25% to 35% of the whole-body protein content. Type-I collagen is the most abundant collagen of the human body. It is present in scar tissue, the end product when tissue heals by repair. It is found in tendons, the endomysium of myofibrils and the organic part of bone.
