Immunohistochemical staining using Collagen Type IV antibody (biotin) (60R-CR005bt)
Anti collagen IV antibody (1:400, 45 min RT) showed strong staining in FFPE sections of human kidney (Left) with strong red staining observed in glomeruli and liver (Right) with strong staining in sinusoids. Staining for both tissues was consistent with a basement membrane distribution. Slides were steamed in 0. 01 M sodium citrate buffer, pH 6. 0 at 99-100 deg C - 20 minutes for antigen retrieval.

| Form & Buffer | Lyophilized from 20mM Potassium Phosphate, pH 7.2, with 150mM Sodium Chloride, 10mg/ml BSA as a stabilizer (Immunoglobulin and Protease free), and 0.01% Sodium Azide added as a preservative. Reconstitute with 0.1ml of deionized water (or equivalent). |
Usage & Assay Information
| Usage Recommendations | ELISA: 1:20,000-1:100,000, IHC: 1:1,000-1:5,000, WB: 1:2,000-1:10,000 |
|---|---|
| Bioactivity | Some class-specific anti-collagens may be specific for three-dimensional epitopes which may result in diminished reactivity with denatured collagen or formalin-fixed, paraffin embedded tissues |
Storage & Safety
| Storage | Ships ambient or refrigerated. Upon receipt store at 4°C while lyophilized. After reconstituted, store at 4°C short term, or aliquot and store at -20°C long term. Avoid repeat freeze-thaws. |

| Biohazard Information | This product contains sodium azide as preservative. Although the amount of sodium azide is very small appropriate care must be taken when handling this product. |
General Information
| Biological Significance | Collagen is a group of naturally occurring proteins found, in nature, exclusively in animals, especially in the flesh and connective tissues of mammals. It is the main component of connective tissue, and is the most abundant protein in mammals, making up about 25% to 35% of the whole-body protein content. Type-IV collagen is a type of collagen found primarily in the basal lamina. The C-terminus domain is not removed in post-translational processing, and the fibers link head-to-head, rather than in parallel. Also, type-IV lacks the regular glycine in every third residue necessary for the tight, collagen helix. |
