General Information

Product Use Monoclonal FN1 antibody specific for Human Fibronectin 1 for use in Immunofluorescence.
Biological Significance Fibronectin is a high-molecular weight glycoprotein of the extracellular matrix that binds to membrane-spanning receptor proteins called integrins. Similar to integrins, fibronectin binds extracellular matrix components such as collagen, fibrin, and heparan sulfate proteoglycans. Fibronectin has numerous functions that ensure the normal functioning of vertebrate organisms. It is involved in cell adhesion, growth, migration, and differentiation. Cellular fibronectin is assembled into the extracellular matrix, an insoluble network that separates and supports the organs and tissues of an organism.
Expression Altered fibronectin expression, degradation, and organization has been associated with a number of pathologies, including cancer and fibrosis.
Implications in Disease Defects in FN1 are the cause of glomerulopathy with fibronectin deposits type 2 (GFND2); also known as familial glomerular nephritis with fibronectin deposits or fibronectin glomerulopathy. GFND is a genetically heterogeneous autosomal dominant disorder characterized clinically by proteinuria, microscopic hematuria, and hypertension that leads to end-stage renal failure in the second to fifth decade of life. Mutations in fibronectin expression, degradation, and organization have been associated with a number of pathologies, including cancer and fibrosis. Fibronectin has been implicated in carcinoma development. In lung carcinoma, fibronectin expression is increased, especially in non-small cell lung carcinoma. The adhesion of lung carcinoma cells to fibronectin enhances tumorigenicity and confers resistance to apoptosis-inducing chemotherapeutic agents.
Sequence Information FN1 is composed of 2386 amino acids with a molecular weight of 262625 Da. There is a DNA binding region at 907 – 1172, 2 fibronectin type-I domains, 2 fibronectin type-II domains and 16 fibronectin type-III domains.
Post-Translational Modifications FN1 forms covalent cross-links mediated by a transglutaminase, such as F13A or TGM2, between a glutamine and the epsilon-amino group of a lysine residue, forming homopolymers and heteropolymers (e.g. fibrinogen-fibronectin, collagen-fibronectin heteropolymers). Phosphorylation sites are present in the extracellular medium. Proteolytic processing produces the C-terminal NC1 peptide, anastellin. FN1 undergoes ubiquitination at Lys1050 and glycosylation at Thr278, Thr279, Ser280, Ser281, Ser283, Thr287, Asn430, Asn528, Asn542, Asn877, Asn1007, Asn1244, Thr1276, Ser1565, Ser1566, Ser1567, Thr1570, Thr1641, Thr1999, Thr2060.
