Flow Cytometric analysis using HLA MHC Class I antibody (10R-6691)
Staining of normal human peripheral blood mononuclear cells either unactivated (left) or 3-day PHA-activated (right) with 0.25 ug of Mouse IgG1 kappa Isotype Control (open histogram) or 0.25 ug of Anti-Human HLA- MHC Class I free chain without beta 2 microglobulin Purified (filled histogram) followed by F (ab'')2 Anti-Mouse IgG (PE). Total viable cells were used for analysis.

General Information
| Biological Significance | The A4 monoclonal antibody reacts with the class I human leukocyte antigen complex in the absence of beta2 microglobulin. There are three class I alpha-chain genes in humans, called HLA-A, -B and -C. Normally, HLA class I proteins are associated non-covalently with beta2 microglobulin. The function of the HLA class I complex is to present peptides derived from intracellular pathogens, including viral pathogens, on the cell surface to antigen specific CD8+ cytotoxic T cells. Class I HLA is expressed by the majority of nucleated cells. |
