HP1 alpha antibody was raised in Rabbit using a KLH-conjugated synthetic peptide encompassing a sequence within the center region of human HP1 alpha as the immunogen
Western blot analysis using HP1 alpha antibody (70R-50897)
Western blot analysis of HP1 alpha expression in HeLa (A); NIH3T3 (B); C6 (C) whole cell lysates.
HP1 alpha antibody
Western blot analysis using HP1 alpha antibody (70R-50897)
Western blot analysis of HP1 alpha expression in HeLa (A); NIH3T3 (B); C6 (C) whole cell lysates.
Immunohistochemical analysis using HP1 alpha antibody (70R-50897)
Immunohistochemical analysis of HP1 alpha staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Immunofluorescent analysis using HP1 alpha antibody (70R-50897)
Immunofluorescent analysis of HP1 alpha staining in NIH3T3 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

General Information
Biological Significance
HP1 alpha is a component of heterochromatin that recognizes and binds histone H3 tails methylated at 'Lys-9' (H3K9me), leading to epigenetic repression. In contrast, it is excluded from chromatin when 'Tyr-41' of histone H3 is phosphorylated (H3Y41ph).
