|Assay Information||1. Prepare a 150ul reaction mixture into a suitable container. Reaction mixture: 72mM Imidazole ( pH 7.5), 7.6mM ADP, 15mM MgCl2, 72mM KCl, 0.2mM Beta-NADH. 2. Incubate for 10 min at 37C. 3. Add 50ul of 20 units/ml LDH, 50ul of 5mM PEP, and incubate for 10 min at 37C. 4. Add 50ul of recombinant PKM2 protein (in 0.1% BSA / PBS) with various concentrations (0.03ug -1ug). 5. Read the decrease in A340nm in kinetic mode for 10 minutes.|

|Biological Significance||PKM2 is an isoenzyme of the glycolytic enzyme pyruvate kinase. This protein catalyzes the production of pyruvate and ATP from phosphoenolpyruvate. PKM2 interacting with Opa proteins, a bacterial outer membrane protein involved in gonococcal adherence to and invasion of human cells, is required for bacterial pathogenesis. It is specifically expressed at high levels in tumor cells, and can be measured in plasma of patients with advanced breast cancer. Recombinant human PKM2, fused to His-tag at N-terminus, was expressed in E. coli and purified by using conventional chromatography.|
