General Information
| Biological Significance | The cyclooxygenase isoenzymes, COX-1 and COX-2, catalyze a key step in the conversion of arachidonate to PGH2 in the formation of prostaglandins, thromboxane, and levuloglandins. Prostaglandinshavecritical functionsin numerous biologic processes, including the regulation of immune function, kidney development, reproductive biology, and gastrointestinal integrity. |

| Expression | The COX1 enzyme is produced constitutively (i.e., gastric mucosa). This functional role for COX1 is consistent with its tissue expression patterns, where nearly all normal tissues were found to express COX1, including kidney, lung, stomach, duodenum, jejunum, ileum, colon, and cecum of rat, dog, Rhesus monkey, and human. |
| Implications in Disease | Epidemiologic, animal, and human data indicate that NSAIDs, inhibitors of cyclooxygenase, are chemopreventive for colon cancer. NSAID use reduces risk for Alzheimer's Disease (AD), with users of these agents having as little as one half the risk of acquiring AD as those not taking NSAIDs. |
